- W2182238595 abstract "While atomic force microscopy technology interrogates biological materials through mechanical interactions, achieving quantitative mechanical information was a challenging task. Mapping full suite of mechanical properties, including modulus, deformation, adhesion and mechanical dissipation, with molecular resolution was proved to be even more difficult. This presentation reports progresses using peak force controlled atomic force microscope to map mechanical properties of broad range of samples including cells, collagen, protein membranes and DNA molecules. The peak interaction force, ranging from 15 pN to 10 �N, was controlled precisely similar to triggered force volume but with over 3 orders of magnitude of higher efficiency and sensitivity of force level. The interaction force control at pN level yield high resolution images while other mechanical data were recorded simultaneously. Using direct peak force control, imaging in fluid also became far easier by elimination of control parameters and cantilever tuning process. The quantitative mechanical data were derived with accurate calibration of cantilever spring constant and tip radius. For elastic modulus measurements a set of bench mark samples from 0.7 MPa to 70 GPa with well controlled properties was used to verify the accuracy quantitatively. Data from bacteriorhodopsin membrane, lamda DNA, collagen and living cells will be discussed as examples of the new peak force controlled mapping method." @default.
