We have recently been investigating the role of scleral papillae in the development of scleral ossicles in the eye of the cmck. This study involved surgical manipulation of the tissues of the chick eye between 61hand 12 days of incubation. Such stages have always been difficult for in. ovo experiments, owing to the fact that after the 4th day of incubation the embryo becomes surrounded by the amnion, the chorion and the aUantois. Wounding these membranes in order to gain access to the embryo results in high mortality and sUIgery must be performed through minute incisions, wmch very severely limits the manipulationsthat can be performed. In OUrexpe.rienee, wounds of a size which cannot heal almost immediately result in the prolonged leakage of extra.embryonic fluid, which floods the outer surface of the chorio·allantoic membrane (CAM), Embryos contained in such eggs usually dIe within 24 h, possibly as a result of disruption of nOJ'mal gaseous exchange through the mem· branos. To Overcome this problem we have devised a meth.adby which to seal incisions in the CAM, thuS preventing leakage of fluid. Our method enableS us to make holes in the CAM large enough for complex surgical manipulations on the eyes of the embryos. The procedure we use is particularly useful for investigating craniofacial development by surgical interference and could also be adapted for studies on other parts of the body of the embryo.
