- W2183651196 abstract "Type 2 diabetes mellitus (DM2) constitutes an increasing worldwide public health problem. This disease is defined by a deficient secretion or peripheral utilization of insulin which drives to severe metabolic alterations and multiple organic damages. Soft tissue infections are common among diabetic patients. DM2 patients may increase peripheral blood leukocytes (PBL) apoptosis due to the persistent hyperglycemia, which may impair the immune response against infections. Therefore, in this work we studied the presence of apoptosis of PBL in DM2 patients with soft tissue infections, and weather there is correlation with the clinical features. The proportion of cells with “spontaneous” or “induced” apoptosis was assessed determining active caspase 3, Fas, and FasL expression by immunohistochemistry, and DNA fragmentation by TUNEL in PBL. We found that patients with DM2 and soft tissue infections showed more induced apoptosis (TUNEL) and FasL expression in PBL. Correlation between induced apoptosis and FasL or glycemia was found, which also correlated with Fas and FasL expression. Correlation between FasL expression and the DOQUI index and blood osmolarity was also found. In this work we demonstrated for the first time an increase in PBL induced apoptosis of DM2 patients with infection, which in part may explain the immumosupression they present. These results suggest a caspase dependent apoptosis mechanism with the participation of Fas and FasL. The correlation between glycemia and Fas and FasL expression suggests that glucose may, directly or indirectly, induce the expression of these molecules, which may explain the induction of apoptosis in DM2 patients" @default.
