Crownsofhumanteeth, eachwithoneexposed window, wereprepared ingroupsoften.Theywereeachpretreated byimmersion individually in4mlofeither (1)clarified wholesaliva for18,72, or168h,(2)dialyzed saliva (3500 MWCO membrane), (3)theflow-through fraction fromaDEAE separation ofwholesaliva (neutral andbasic proteins), (4)theeluted fraction ofaDEAEseparation ofwholesaliva (anionic proteins), or(5) acombination ofsalivary lipids andtheDEAEflow-through fraction ofwholesaliva (neutral andbasic proteins). Control groupsweregroup6with no pretreatment, group7pretreated for168hinaborate buffer (5mmol/l), and group8pretreated inamineral solution containing calcium (0.7 mmol/1) andphosphate (2.6 mmolfI). Thecrownswerethen demineralized for7dinvitro (0.1 mol/I acetate, 1 mmol/I Caandphosphate, pH5.0) toproduce artificial caries-like lesions. Lesions wereassessed bycross-sectional microhardness profiles, andmineral loss (AZ,Rim xvol%mineral) calculated. Mineral loss (AZ)values decreased linearly withthesquarerootoftimeofpretreatment bywholesaliva, confirning atime-dependent protective effect ofsalivary pellicle against demineralization ofenamel. Pretreatments (168h)bywholesaliva (group 1), dialyzed saliva (group 2),andlipid/'flow through proteins (group 5)gaveequivalent protection (approximately 55%). However, no protection was provided byDEAE-separated protein fractions (nolipid present) orbythemineral alone. The protection ofsurface enamel against demineralization appearstobegiven byacombination of specifically adsorbed salivary lipids andproteins." @default.
