Moreover, gA andgBofHSV-1 crossreact inserologic tests with thecorrespond- ingglycoproteins ofHSV-2. Inthis paper, wereport onthereac- tivities ofgAandgBofHSV-1 andHSV-2 with 24independently derived monoclonal antibodies reactive with these antigens. Our results showthefollowing: (i) Electrophoretic mobilities ofHSV- 1andHSV-2 glycoproteins gAandgBmadeinHEp-2 cells are significantly less than those madeinVero cells. (ii) All monoclonal antibodies precipitated both gAandgBmadeininfected HEp-2 cells. These include 12monoclonal antibodies that neutralized vi- rusand12that didnot. (iii) HSV-2 glycoproteins gAandgBmade inHEp-2cells contain type-specific domains. However, mono- clonal antibodies produced byoneclone directed tothese domains didnotreact with glycoproteins madeinVerocells. (iv) Lysates ofinfected Verocells contain three lower molecular weight poly- peptides that also reacted with monoclonal antibodies directed to glycoproteins gAandgB.These polypeptides arevirus specific inasmuch asthose specified byHSV-1differ inelectrophoretic mobilities from those specified byHSV-2. Although gAandgBcanbedifferentiated onthebasis oftheir electrophoretic mobilities inNaDodSOpolyacryla- midegels (1), both react with antisera prepared against each of theglycoproteins (12). Inthis paper, wereport theresults of analyses ofHSV-1andHSV-2glycoproteins gAandgBmade ininfected humanepidermoid carcinoma 2(HEp-2) andAfrican green monkey kidney (Vero) cells with monoclonal antibodies reactive with these glycoproteins. MATERIALS ANDMETHODS Viruses. Theisolation andproperties ofHSV-1(F) andHSV- 2(G) have beendescribed (13, 14). Virus stocks andviral anti- gens wereprepared inHEp-2andinVerocells asdescribed (1, 15). Monoclonal Antibodies. Hybridoma cell lines werederived fromfusion ofBALB/c MOPC 21NS-1myeloma cells with spleen cells from miceimmunized with HSV-1 orHSV-2. The procedures for theproduction andselection ofhybridomas and theimmunologic properties ofmonoclonal antibodies from HC1 andHD1hybridoma cell lines have been described (16). Ascites fluids containing monoclonal antibodies wereproduced byin- traperitoneal injections ofhybridoma cells into miceprimed with pristane (Aldrich). Theneutralization tests weredoneas described (16). Preparation ofRadiolabeled Antigen Extracts. Cells in- fected with 10-20 plaque-forming units ofstock virus percell werelabeled from 6hrafter infection with (3S)methionine (50 ,ACi/ml, >400Ci/mmol, 1Ci= 3.7X 1010 becquerels; New England Nuclear) inmedium containing 1/10 thenormal con- centration ofmethionine.
