W2186783665 abstract "HER2�geneamplificationininvasivebreastcancerisarobustpredictivemarkerforresponsetotranstuzumab� therapy.�Thisstudywasundertakentomeasureconcordancebetweenimmunohistochemistry�(IHC)�andFISH� forHER2�geneamplificationininvasivebreasttumors,�aswellasthepresenceofpolysomy�17�andpossible� correlationwithdemographicsandhistopathologicalvariables,�includingERandPRpositivity.�Atotalof�425� casesofinfiltratingcarcinomaofbreast�(99%�IDC-NOS)�werestudied.�HER2�overexpressionwastestedbyIHC� andFISHmethods.�AssociationbetweenIHCandFISHinbothsubsetswascalculatedbyamplificationratio� includingpolysomy�17.��Outof�425�specimens,�128�(30%)�werepositiveforHER2�amplificationbyFISHtest,� whereasonly�78�(24%)�tumorswith�2+�expressionshowedamplification.� Incontrast,�39�(74%)�demonstrated�3+� IHCscoreandHER2�geneamplification.��Thehistologicalvariablesincludingtumorsize,�tumortype,�andlymph� nodeinvolvementdidnotinfluencetheoutcomeofFISHanalysis.�TheERandPRstatusshowedsignificantly� greaterpositivityinpatientsnegativeforHER2�amplification.� Polysomy�17�wasdetectedin�23.7%�patients� andwaspositivelyassociatedwithERandPRexpression�(P=�<0.05).�Ourstudyshowedaconcordanceof�24%� between�2+�IHCandFISHamplification,� whilein�3+�IHCcasestheconcordancewas�74%.�Significantlinksof� HER2�amplificationwasseenwithERandPRnegativityandhighertumorgrade.�Inaddition,�non-significant� correlationswerenotedwithothervariablesliketumortype,�sizeandlymphnodestatus." @default.
