- W2186937807 abstract "Switchgrass (Panicum virgatum L.) shows potential for use in the biofuels industry, but establishment can be unreliable due to seed dormancy. Typically, after seeding, it may take up to three years to reach a proper stand for maximum yield. Scarification, stratification, and/or the use of chemicals have been shown to facilitate the breaking of dormancy and increase germination. The current protocol for testing seeds requires up to 28 days and generally results in a 5070 % germination rate, with positive tetrazolium tests in non-germinated seeds showing 20-40% dormancy. Initial experiments, with Sunburst and Pathfinder cultivars, examined seed weight increases during imbibition. These data showed that water enters floral parts but dormancy still occurs. All seeds incurred an initial increase in water weight, but only germinating seeds demonstrated the typical triphasic pattern of imbibition. This indicates that water uptake does not control dormancy and suggests that the floral parts may block the exit of chemical inhibitors. Scarification, including stripping the palea and lemma (floral parts) from the seed, typically results in higher germination rates in this species. Abscisic acid (ABA) is known to be involved in seed dormancy in other plant species and may be a chemical inhibitor in switchgrass. Immunoassays were performed to examine levels of ABA in the seeds. ABA was detectable in both germinating and non-germinating switchgrass seeds and was found to have been released from both of these seed types into the external media. These data suggest a role for ABA in maintenance of dormancy in this species, but do not suggest that ABA depletion completely controls switchgrass seed germination. Scarification can also induce ethylene production, which is often associated with the breaking of seed dormancy. Treatment of Sunburst seeds with ethephon (an ethylene promoter) significantly increased germination, but was much less effective in the Pathfinder seeds. These data indicate that ethylene production is involved in the normal breaking of dormancy. Tissue culture of excised embryos confirmed that the dormancy is not a function of the embryo, but involves inhibitory compounds found in the other components of the seeds, and suggests that germination is stimulated by promoter compounds that overcome inhibition by ABA." @default.
