- W2187715908 abstract "Introduction: β - lactamase enzymes are CTX - M enzymes that hydrolyze ceph alo t in e and c efaloridin e better tha n penicillin, and also preferentially hydrolyze c efotaxime over c eftazidime . The aim of the study was to investigate the prevalence of ESLB - producing E. coli , and molecular evaluation of CTX - M β - lactamases using Multiplex in E. coli strains . Materials and methods : A total of 200 E. c oli isolates were collected from clinical specimens and the antibiotic susceptibility of strains was determined using Disk Diffusion method. The p roduction of ESBL enzymes was determined using Combined Disk , C efotaxime and C eftazidime with and without Clav ul anic Acid. Also, MIC of Cefta zidime and Cefotaxime was determined w ith and without Clavulanic Acid using Agar Dilution method . Finally , t he presence of CTX - M genes was detected by specific primers using Multiplex PCR method . Findings : The drug resistance of separated i solate s was determined in relation to 14 anti - Antibiotics. In the C ombined Disk method , increase in inhibition zone diameter by ≥ 5mm against c efta zidime and cefotaxime with and without Clavulanic A cid as ESBL producers were identified. There were 155 isolates (77.5 % ) in C ombined Disk method as ESBL - producing E. coli . Among these ESLB - producing isolates , 66 ESBL samples contained MIC CAZ = 16 - 32 (µg/ml) , and 24 samples contained 64 to 512. Also, 48 samples contained MIC CTX =4 - 8 and 97 samples contained 16 to 512 for CTX. 4/57% of the se isolates in Multiplex PCR method were CTX - M - producer s . Conclusion : The results show that the production of β - lactamase ESBL enzymes in the strains is high (77.5%) , which is confirmed by A gar D ilution and Combined Disk methods. PCR method shows higher prevalence of CTX - M enzyme . Therefore , appropriate treatment is recommended in the country to prevent the spread of ESBL - producing Escherichia coli strains .
