- W2188152566 abstract "When determined undertheusualcondi- tionsofanexcess ofligand overprotein, theconcentration ofcyclic AMP necessary toactivate purepreparations of cyclic AMP-dependentprotein kinase(EC2.7.1.37; ATP:- proteinphosphotransferase) half-maximally isin the rangeof0.2-0.3 uM whencaseinorglycogen synthetase is usedasthesubstrate, i.e., essentially thesameasthecon- centration ofthenucleotide thatisfoundinresting skele- talmuscle.Theapparentdissociation constant forcyclic AMP boundtotheprotein kinaseisalsoabout0.2-0.3 ,M whenmeasuredundersimilar conditions. Theconcentra- tionoftheproteinkinaseinmuscleisrelatively high (0.23 MM),however, andundertheseconditions theappar.. entactivation constantoftheenzymeforcyclic AMP is raised sothatanincrease incyclic AMP levels inthetissue wouldcausea concomitant increase inproteinkinase activity overa widerangeofnucleotide concentration. Asaresult, itisunnecessary toinvokecompartmentaliza- tionofcyclic AMP toexplain how itcancontrol protein kinaseactivity invivo.Anotherfactor thatmay increase theeffectiveness ofchangesincyclic AMP concentration istheheat-stable protein inhibitor ofprotein kinasethat may function toinhibit theactivity ofnearlyallthepro- teinkinasecatalytic subunitdissociated bybasalconcen- trations ofcyclic AMP. Finally, thenearequitybetween theconcentration ofcyclic AMP bindingsitesand the liganditself provides a potential mechanismwhereby agentscanaffect thetotalcyclic AMP contentwithout directly affecting adenylate cyclase, cyclic AMP phospho- diesterase, orcyclic AMP transport. Itisnowwidely accepted thatmanyhormonal effects are mediated, atleast inpart, bychanges intheconcentration of cyclic AMP inthecell (1). Ithasfurther beenfound thata numberoftheactions ofcyclic AMP inmammalian tissues takeplace through theinteraction ofcyclic AMP withthe regulatory subunit ofcyclic AMP-dependent protein kinase (EC2.7.1.37; ATP:protein phosphotransferase)" @default.
