- W2188528300 abstract "and intrinsic protein oftheoutermembranewere stable, some oftheregularly arranged surface protein, thea-protein, was lost intothegrowth medium.Only newlysynthesized a-protein was lost. Thepeptidoglycan ofthemureinlayer was also labile. Selective inhibition oftheformation ofindividual components of thecell envelope withpenicillin, chloramphenicol, andbacitracin showedthat incorporation ofprotein intotheoutermembranerequired thesimultaneous formation ofcomplete lipopolysaccharide. Theconversewas nottrue:protein synthesis was notrequired forlipopolysaccharide incorporation. Formation of theoutermembraneandthemureinlayer proceeded independently. Thecell envelope ofbacteria isacomplex but well-defined structure. Gram-negative bacteria naveanenvelope thatismultilayered, with plasmamembrane, murein, outermembrane, sometimes asurface protein, andpossibly also intermediate layers (7). Theformation ofthe cell envelope therefore requires precise chemicalandtopological control andthereby provides avaluable experimental system forinvestigatingregulation ofsynthesis ina multicomponentstructure. Although aconsiderable amountofworkhas beendoneontheregulation ofplasmamembraneformation (15), less isknownabout the otherlayers. Components oftheoutermembrane aresynthesized attheplasma membrane (3,19), butthemechanism whereby theyare incorporated incontrolled fashion isnotunderstood. Oneofthecomponents oftheouter membraneisaregularly arranged surface protein. Thesynthesis andturnover ofthisprotein is considered inparticular detail inthispaper. Thecharacteristics ofitsformation provide a mechanism wherebythecellisalwayscompletely covered withsurface protein. Thisindicates thatthesurface protein hasanimportant biological function. Since theoutermembraneofAcinetobacter sp.strain 199Acanbeprepared (31) andindividual components canbeisolated, itispossible touseittofollow thesynthesis andturnover of eachcomponent andtoexamine their dependenceononeanother andonthemureinlayer of thecell envelope.
