- W2189012004 abstract "Members ofthegenus Actinomyces arepredominant primary colonizers oftheoral cavity andplay animportant role ininitiating plaque development. These bacteria haveevolved unique mechanisms that favor colonization andpersistence in this micro-environment. Theexpression ofcell-surface fimbriae iscorrelated with theability ofthese bacteria toadhere tospe- cific receptors onthetooth andmucosal surfaces, andtointeract withother plaque bacteria. Theelaboration ofsialidase is thought toenhance fimbriae-mediated adherence byunmasking thefimbrial receptors onmammalian cells. Thepresence of certain cell-associated orextracellular enzymes, including those involved insucrose orureametabolism, mayprovide the meansforthese bacteria tothrive under conditions whenother growth nutrients arenotavailable. Moreover, these enzyme activities mayinfluence thedistribution ofother plaque bacteria andpromote selection forActinomyces spp. incertain ecolog- ical niches. Therecent development ofagenetic transfer system for Actinomyces spp. hasallowed forstudies theresults ofwhich demonstrate theexistence ofmultiple genes involved infimbriae synthesis andfunction, andfacilitated theconstruction of allelic replacement mutants ateachgenelocus. Analyses ofthese mutants haverevealed adirect correlation between thesyn- thesis ofassembled fimbriae andtheobserved adherence properties. Further genetic analysis ofthevarious enzyme activities detected fromstrains ofActinomyces should allow foranassessment oftherole ofthese components inmicrobial ecology, and their contribution totheoverall success ofActinomyces spp. asaprimary colonizer andakeyplayer inoral health anddisease." @default.
