Furthermore, thesame cells thathadmigrated toward oneattractant wereabletorespond toother chemotaxins. Wealso found thatasingle attractant attracted asmanycells asa combination oftwoorthree attractants. Calculations fromthese datashowed that atleast 75%ofthemigrating monocytes havedifferent receptors forall three attractants. Macrophages playanimportant roleinthe immunological response toneoplasms andare foundatthesite ofprimary andtransplanted tumors. Since peripheral blood monocytes are precursors ofthese effector macrophages, the ability ofmonocytes tomigrate maybeessential foraccumulation attumorsites. Monocytes from cancer patients haveabnormal chemotaxis in vitro. Decreased monocyte chemotaxis wasassociated withpoorprognosis (14); theabnormal response wasreversed bysurgical removal ofthe tumor(13). Boetcher andLeonard showeddifferences inmonocyte response totwodifferent chemotactic agents, lymphocyte derived-chemotactic factor (LDCF)andactivated serum(6). Only50%ofcancerpatients withimpaired responses toLDCFalso hadreduced responses to activated serum.Thesefindings suggest that either there arespecific subpopulations ofmigrating monocytes withdifferent chemotaxin receptors orthatspecific receptors inahomogeneouspopulation withmultiple receptors on eachcell wereblocked. Toresolve these alternatives, weexamined subpopulations ofmigratingmonocytes innormal subjects. We showby various chemotaxis experiments using 10-,umthick polycarbonate filters thatthere isonly one subpopulation amongallmonocytes that isable tomigrate invitro, andthatmostofthese cells respond to allthreechemotactic stimuli tested." @default.
