- W219181389 abstract "This chapter describes methods for identifying the characteristics of mononuclear phagocytes from different tissues. The preparations of all cell suspensions are made by spinning down the cells onto a glass cover slip. For this purpose, a round cover slip with a diameter of 12 mm is placed in a flat-bottomed centrifuge tube. Then, 3 ml of phosphate-buffered saline containing 5% newborn calf serum is pipetted into the tube. When enzyme digestion is used, as for liver and lung tissue, the streptomycin is replaced by 20 μg/ml gentamycin. After incubation, the cover slips are washed with serum-free medium, removed from the tube, quickly air-dried, and finally mounted cell side up on slides for staining. In Giemsa-stained direct preparations of bone marrow, blood, and peritoneal fluid, the mononuclear phagocytes of the different species show great similarity but their behavior on a glass surface differs in some respects. An important question to be considered is how high the percentage of marker-bearing cells in a population must be before that population can be called a mononuclear phagocyte population. As almost none of the currently used markers give 100% positivity, 90% seems to be acceptable for the present." @default.
