- W2200617064 abstract "In 1952, the Belgian mycologist Vanbreuseghem developed a selective method for isolating kera-tinophilic fungi from soil, which has been widely known as the hair-baiting technique. By means of this technique, he obtained from soil in Belgium a keratinophilic fungus, hitherto unknown, which he gave the name Keratinomyces ajelloi. In 1953, Ajello isolated a known dermatophyte, or Microsporum gypseum, from soil in America. Since then, by the same method, both species of keratinophilic fungi have been isolated many times from soil all over the world. Recently, other keratinophilic fungi, such as Trichophyton terrestre, T. mentagrophytes, T. quinckeaum, and M. cookei, were isolated from soil in some countries. In Japan, however, only one species of keratinophilic fungi, or K. ajelloi, has been isolated from soil up to the present. This technique was described in detail in the papers of Vanbreuseghem and Ajello It is outlined as follows. Sterile Petri dishes are half-filled with soil specimens, moistened with a certain quantity of sterile water, and baited by placing short tufts of sterile hair on soil. If a keratinophilic fungus inhabits soil, it grows soon on the hair bait and its mycelium becomes visible with a naked eye. Since human hair was used by Vanbreuseghem as bait in his technique, it has been used routinely by later workers. Dawson and Gentles employed such keratinaceous material as horse mane and tail hair, child's hair, shreds of cow horn, and feathers other than human hair. They observed that K. ajelloi, isolated from soil, produced its perfect stage, or cleistothecia, on horse hair and child's hair, Later, they studied cleistothecia of certain species of keratinophilic fungi by the same method and by means of a soil culture. The authors were planning a survey on the presence of keratinophilic fungi in soil at areas which are frequented by animals. Prior to this survey, they wanted to know whether animal hair could be used as bait, as well as human hair, in the hair-baiting technique. As a result, it was proved that the hair of any common species of mammals could be used as bait. Therefore, they began their soil survey by this technique, using horse hair as well as human hair." @default.
