- W2205574908 abstract "Abstract Almost two decades ago, three novel members of the tumor necrosis factor (TNF) and TNF-receptor (TNFR) superfamily, namely the receptor activator of NFκB (RANK), the ligand RANKL, and the decoy receptor osteoprotegerin (OPG) were discovered 1–7. In analogy to various other members of the family of TNF- and TNFR molecules, the initial function of RANK, RANKL, and OPG was thought to be important for immune responses. However, it turned out that RANK and RANKL are the key unit controlling bone turnover, via regulation of osteoclast differentiation and activation. Throughout the years, many more functions of RANK and RANKL have been discovered, beyond bone physiology. RANK and RANKL are the key molecules in mammary gland development during pregnancy, and control the onset of hormone-induced breast cancer, as well as metastatic spread to bone tissue. Moreover, RANK and RANKL are critical for the establishment of central immune tolerance, fever responses in the brain, and hair renewal. In this chapter, we will discuss the diverse functions of RANK and RANKL, ranging from bone physiology, immune regulation, and mammary gland biology" @default.
