- W2205854605 abstract "Publisher Summary This chapter discusses the antibody approach to membrane architecture. Antibodies are valuable tools for biochemical investigations, because their reaction with soluble antigens is highly specific, fast, and strong. Precipitation and inhibition of enzymes are useful for the detection of the interaction of antigen and antibodies. When applied to membranes, the immunological methods partially have to be revised, but additional conclusions can be drawn concerning the location of the antigens as constituents of membranes. This approach to the architecture of the chloroplast thylakoid membrane system has been reviewed together with considerations on the correlation of structure and function, especially the asymmetric distribution of components perpendicular to the thylakoid surface, their distribution along this membrane system, and the action of inhibiting antibodies on photosynthetic electron transport. In this chapter, the basis of the approach, the experimental possibilities, the conclusion patterns, and the inherent limitations are considered. A complementary treatise on the application of antibodies to studies on the mitochondrial membrane is presented. Antibodies as membrane surface probes are introduced. A discussion on experimental possibilities, patterns of conclusions, experimental procedures, reproducibility and limitations is also presented in the chapter." @default.
