- W2212129211 abstract "This chapter discusses the use of synthetic peptides as a diagnostic tool in virology. Synthetic peptides have been considered to be valuable tools for viral laboratory diagnosis as they provide a uniform, chemically well-defined antigen for antibody analysis, diminishing inter- and intra-assay variation. Synthetic peptides can also be used to define biologically important epitopes that can then be utilized in vaccine development or measured for diagnostic purposes. Several empirical methods have been developed that are based on the properties of amino acid residues to determine the presumed location of continuous B cell epitopes in native proteins. Even though assays employing synthetic peptides in solution are regarded as optimal for the characterization of epitopes, most diagnostic tests use peptides bound to solid phases. Human immunodeficiency virus has been particularly rewarding for applications using synthetic peptides for laboratory diagnosis. Handling of infectious virus in the laboratory is hazardous, making large-scale production of viral antigens unattractive. Intensive basic research has provided detailed information about the structure and function of different parts of the virion. Furthermore, immunologically dominant structures have been identified that can be mimicked by linear synthetic peptides. By using these peptides, tests for HIV antibodies have been developed that have close to 100% sensitivity and specificity." @default.
