- W2215707863 abstract "目的探讨靶向CCAAT／增强子结合蛋白-β（C／EBPβ）基因沉默对肝癌细胞SMMC-7721增殖、凋亡、迁移的影响。方法设计C／EBP-β及eGFP、shRNA序列，装入pLKO．1-TRC质粒，进行慢病毒包装，感染肝癌SMMC-7721细胞，嘌呤霉素筛选并培养稳定干扰细胞株，cellcountingkit-8（CCK-8试剂盒）法、transwell细胞迁移实验分别检测沉默C／EBP-β基因对SMMC-7721细胞增殖和迁移能力的影响，蛋白印迹（Westernblot）检测干扰效率及沉默C／EBP—p基因对SMMC-7721细胞的凋亡相关蛋白影响。结果沉默C／EBP．B基因的SMMC-7721．pLKO—sh—C／EBP—p肿瘤细胞的增殖速度明显提高（相对增值率136．8％，P〈0．05），细胞迁移能力也明显增强（迁移抑制率为-292．3％，P〈0．05），且细胞周期蛋白cyclinB1的表达增强（P〈0．05），抗凋亡蛋白Bcl-2表达增强（P〈0．05），凋亡相关蛋白Bax的表达下降（P〈0．05）。结论沉默C／EBP—β对肝癌细胞SMMC-7721的增殖、迁移有促进作用，对其凋亡有抑制作用。" @default.
