- W221971379 abstract "This chapter provides an overview of the neural release induced by calcium ionophores. Both X-537A and A 23187 serve as carriers of Ca2+ and are able to initiate a wide variety of Ca2+-dependent events in a fashion independent of specific receptor activation. However, neither ionophore shows absolute selectivity for Ca2+ and both can complex and transport other divalent and monovalent cations. In addition to their ability to complex and transport inorganic cations, both X-537A and A 23187 can complex a number of organic cations. The actions of both X-537A and A-23187 have been examined in a number of brain synaptosome preparations derived from both whole brain and discrete brain areas. Both X-537A and A 23187 release catecholamines by at least two distinct processes. The importance of these two actions of X-537A are dependent upon ionophore concentration as X-537A forms 1:1 complexes with catecholamines but 2:1 complexes with calcium. In the absence of such enhanced Ca2+ entry, the normal cellular Ca2+ sequestering/pumping processes are able to maintain a low intracellular Ca2+ concentration despite the ionophore-induced entry of Ca2+." @default.
