Le Chatelier's principle governs the behavior of substances under high pressure. The technique for high-pressure measurements is used in this chapter as a method to investigate enzyme structure. The classification of hydrostatic pressures into low pressure and high pressure is significant. Part of its significance is technical and, in part, is fundamental to the relationship between enzyme structure and pressure. Most of the equipment described in the chapter is constructed and tested in our laboratory; in a few cases, literature descriptions of the apparatus have been relied on. Morita recently gave a good account of the instruments and methods employed by various investigators for studying the effects of pressure (generally below 1000 atm) on biological systems. Hydrostatic pressure equipment designed by various investigators is also listed by Morita." @default.
