- W2238577032 abstract "1841 Objectives For regional quantification of brain PET data, defining the volumes of interest (VOIs) by hand is still considered as the gold standard. Since this manual technique is time-consuming and definer-dependent, a multitude of software tools for the automatic definition of neuroanatomical structures emerged over the last years. The objective of the project was to systematically compare three of the most commonly used tools (PMOD, Hermes BRASS, FreeSurfer) against the conventional method in case of β-amyloid PET data. Methods Individual T1 3D MPRAGE MRI and 70-90min p.i. summed florbetaben β-amyloid brain PET data of 10 patients with probable Alzheimer’s disease (AD) and 10 age-matched healthy volunteers (HVs) were analyzed. VOIs were defined manually as well as automated using PMOD, Hermes BRASS and FreeSurfer. The standardized uptake value ratios (SUVRs) with cerebellar cortex as reference region were obtained for each VOI. Results The neocortical composite SUVRs of the manually defined VOIs were significantly higher for the ADs vs. HVs (p=0.01; Cohen’s d=1.53). The automated methods showed similar group discriminations (PMOD: p=0.004, d=1.87; BRASS: p=0.013, d=1.47; FreeSurfer: p=0.028, d=1.49). Regional SUVRs of the automated and conventional procedures were in most cases significantly correlated, but with variable degree depending on the brain region and software approach (r=-0.28 to r=0.96, p=0.23 to p Conclusions Automated VOI definition tools have great potential to substitute the manually standard procedure in β-amyloid PET data analysis.
