- W2243373254 abstract "In the work presented here, the role of matrix metalloproteinase 19 (MMP19) was studied in inflammatory reactions of the skin. Mice deficient for MMP19 were used to perform two cutaneous inflammation models. In the first model of irritant dermatitis (ICD) inflammatory reactions are independend of T lymphocytes (T cells), whereas the second model of contact hypersensitivity (CHS) requires hapten-specific T cells and thus adaptive immunity. In both models, ICD and CHS, MMP19-deficient mice (MMP19 -/-) exhibit strongly reduced skin inflammation, indicated by minimal edema formation, limited proliferation of keratinocytes, and reduced recruitment of inflammatory cells to the affected site. In the inflamed tissue, MMP19 -/- mice exhibit decreased numbers of T lymphocytes. During CHS, MMP19 -/- mice respond with reduced secretion of proinflammatory cytokines such as I-TAC, lymphotactin, and TCA-3, which act mainly as chemoattractant for T lymphocytes and thus contribute to reduced cellular influx. Moreover, MMP19-deficient mice show diminished activation of CD8-positive T cells, however, accurate T cell activation is essential for the elicitation of hypersensitivity inflammation.Under non-inflammatory conditions MMP19 -/- mice exhibit defective development of T cells that becomes apparent in reduced proliferation and maturation of thymocytes. MMP19-deficient mice show also reduced T cell populations in the blood, pointing towards limited emigration of mature T cells from thymus into the blood. Migration of cells to sites of inflammation requires proteolytic processing of connective tissue components. Since MMP19 is able to cleave several components of extracellular matrix (ECM) proteins such as collagen type IV, fibronectin, laminin, and nidogen, it is proposed that the lack of MMP19 activity contributes to inhibited cell migration. Employing MMP19-deficient macrophages, reduced transmigratory capacity was confirmed in vitro and in vivo.Altogether, MMP19 plays a critical role in inflammatory reactions of the skin, where MMP19 stimulate cytokine production, cell proliferation and migration. Additionally, MMP19 impacts thymocyte development and probably T cell emigration processes into periphery." @default.
