- W2247170256 abstract "A form of acute respiratory failure was produced in rats to study hypoxemia resulting from lung disease and response to drugs. Graded lung damage was induced in Wistar rats by exposure to oxygen at a pressure of one atmosphere for intervals varying between 50 and 65 hours. Abrupt removal from oxygen to air then resulted in acute respiratory distress. In adults rats, the duration of oxygen exposure was related to increasing severity of respiratory difficulty and shortened survival. Respiratory failure did not develop in newborn rats even after 9 days in oxygen, but localized edema and atelectasis appeared in the lungs after 3 days. Rats dying in oxygen showed progressive hypoxemia and terminal hypercapnia and acidemia. Rats dying in air after induction of lung disease by oxygen often hyperventilated and showed alkalemia before the terminal phase of hypoxemia, acidosis, and death. Benzolamide, a renal carbonic anhydrase inhibitor, was given 4 hours before the end of oxygen exposure to produce mild metabolic acidosis. This treatment resulted in increased survival during the subsequent exposure to air and minimized the alkalemia associated with hyperventilation. The experiments showed that oxygen at atmospheric pressure can be combined with air exposure after development of graded amounts of lung damage for convenient study of acute respiratory failure and demonstrated the response to benzolamide in the hypoxemia of lung disease." @default.
