• W2249437045 abstract "Abstract: Introduction: Cell replacement therapy is a promising approach for the treatment of cardiac diseases, but is challenged by a limited supply of appropriate cells. The main purpose of this study is to fabricate a viable tissue construct for cardiac transplantation using skeletal myoblasts (SM). Methods: SM were isolated from neonatal mice and expanded in cell culture. The identity and purity of the cells was confirmed by FACS and western blot analysis. SM from cell culture expansion were seeded into engineered tissue constructs consisting of collagen and basement membrane matrix. The constructs showed sufficient rigidity for mechanical preconditioning and transplantation to infarcted murine hearts. Results: SM were isolated and expanded at high purity (97,1% ± 0,9 desmin positive cells) and expressed myoblast specific markers. Differentiation to excitable and spontaneously contracting myotubes was observed after 7 days of culture in serum reduced media. After seeding into tissue constructs and mechanical preconditioning SM retained their potential for functional integration (Cx43) and formed an artificial tissue which was viable and excitable in vitro. After transplantation these constructs remained viable and may have functionally integrated into the host tissue." @default.
