- W2251840948 abstract "The Cryptococcus neoformans species Complex includes the Cryptococcus neoformans and Cryptococcus gattii yeasts. These basidiomycete yeasts can cause disease in immunocompromised and immunocompetent patients, respectively. Notably in fungus, glicosylphosphatidilinositol-anchored proteins (GPI-Ps) are involved in different aspects of host-pathogen interaction, such as host cells adhesion and invasion, host immune response modulation and evasion and pathogenesis. In this work, we performed a systematic, genome-wide in silico identification of C. neoformans complex species GPI-Ps. A set of prediction tools was apllied for the screening of 6967 and 6210 predicted protein sequences from C. neoformans var. grubii (H99 strain) an C. gattii (R265 strain), respectively. The sequences were retrieved from the Broad Institute genome database (http://www.broadinstitute.org/science/data). The identification of putative GPI-Ps was based on the following criteria: i) the presence of an N-terminal signal peptide for secretion; ii) the presence of a C-terminal GPI-attachment site and iii) absence of internal transmembrane domains. A total of 63 putative GPI-Ps were identified in C. neoformans var. grubii and 47 proteins were identified as GPI-P in C. gattii. Proteins previously described as GPI-anchored, such as Phospholipase B1, Endo1,3--glucanases, Chitin deacetylases and Glyoxaloxidases were identified in both species C. neoformans var grubii and C. gattii. The proteins identified could be classified in several functional categories, especially those involved in cell wall biogenesis and remodeling. About 70% of the GPI-Ps identified have unknown functions. Many of the putative GPI-Ps do not display conserved domains, but some possess have fungal orthologs homologs while others are currently unique to specie. The GPI-P identification in Cryptococcus neoformans species complex is of extreme importance for elucidating aspects related to the pathogenesis and will serve as a model for others fungal diseases.
