- W2254328153 abstract "309 Objectives To evaluate 68Ga-NOTA-UBI 29-41 (68Ga-UBI) for differentiation between bacterial infection (S.aureus) and sterile inflammation using PET/CT, biodistribution, and microbiology methods. Methods Three groups of mice were used for infection and sterile inflammation, compared with a healthy control group. Infection was induced by i.m inoculation of 0,1mL of S.aureus (1.2 x109 cfu/mL) in the thight muscle while inflammation was induced with heat shock treated S. aureus. After 24 hours, four mice per group, were iv injected with 68Ga-UBI and sacrificed after 30, 60 and 120 min. PET/CT dynamic studies (120 min) were performed in a Quad-APD based camera (LYSO/LGSO). Infected or inflamed muscle were dissected, homogenized, diluted 10-1 to 10-4, plated on TSA and incubated 24-48 h to confirm the presence of infection and correlate the specific uptake of 68Ga-UBI. In vitro binding of 68Ga-UBI to St. aureus was evaluated with increasing amounts of microorganism (9,14x106 to 1,17x109 cfu in 0,1 mL) incubated 1h at 40C and followed by centrifugation. Results High renal excretion and rapid blood clearance were observed. In vitro binding was higher than 95% for 5x108 cfu St. aureus in 0,1 mL.
