- W2256964811 abstract "The human gastrointestinal tract contains a diverse microbialcommunity. Among them lactobacilli are important members of thenatural intestinal microbiota playing a beneficial role inbalancing of the intestinal environment. A collection of 51Lactobacillus isolates originating from children intestinalmucous tissues has been studied. Analysed biopsy samples wereretrieved during a routine endoscopic examination of differentparts of the large intestine in patients revealing variousdisorders. A combined approach, which included automatedribotyping and MALDI-TOF MS analysis, was applied foridentification of isolated Lactobacillus spp. in order tocompare the discriminatory power of these methods. Automatedribotyping using the RiboPrinter Microbial CharacterizationSystem (DuPont Qualicon) assigned 38 lactobacilli to thespecies level: Lactobacillus salivarius (29 isolates),Lactobacillus paracasei subsp. paracasei (7) and Lactobacillusrhamnosus (2). The remaining 13 isolates were not identifieddue to low degree of similarity (85% threshold) with thereference ribotype patterns. Additional cluster analysis of theribotype patterns was performed by BioNumerics v. 6.0 software(Applied-Maths) and enabled to assign 10 isolates as L.salivarius. Three remaining strains were not identified by theRiboPrinter/BioNumerics approach. MALDI-TOF MS analysis wasperformed on an Ultraflex III instrument (Bruker Daltonik)according to a standard sample preparation protocol of BrukerDaltonik. MALDI-TOF mass spectra were subjected to numericalanalysis (BioTyper 1.1 software, Bruker Daltonik), whichrevealed the formation of a cluster harbouring all 39 isolatesidentified as L. salivarius and identified all L. paracaseiiand L. rhamnosus isolates being separated in species-specificclusters in agreement with the ribotyping results. Threeisolates unidentified with the RiboPrinter system were assignedas Lactobacillus mucosae by the MALDI-TOF MS. In conclusion,the MALDI-TOF MS enabled reliable identification of allanalysed Lactobacillus isolates to the species level. Incontrast, automatic identification performed by the RiboPrintersystem did not identify 10 L. salivarius and all L. mucosaeisolates.
