- W2264758518 abstract "PNE is a rare neurological disease in the horse, which clinically leads to a cauda equina syndrome and paralysis of one or more cranial nerves. The etiology and pathogenesis of PNE are unknown, but there is a suspicion of an immune response against myelin in PNE, as it occurs in experimental allergic neuritis (EAN) in laboratory animals and the human Guillain Barre Syndrome (GBS)1. In a previous study it was determined with immunohistochemistry that lymphocytes present in the lesions of horses affected with PNE consisted of B-cells and T-cells2.The objective of this second study about the inflammatory infiltrates in PNE is to determine which type of T-cells is present in affected horses, since this could lead to a better understanding of the disease.The hypothesis of this study is that the group of T-cells exists of CD4 positive T-cells, as it is in EAN and GBS3. Three horses with a strong suspicion of PNE were admitted to the Faculty of veterinary medicine of Hannover and finally euthanized 4 months, 12 days and 9 days respectively after the onset of the disease. A final diagnosis of PNE was made on the post-mortem and on histology. Tissue samples of the cauda equina were frozen in liquid nitrogen in a capsule with cryoblock (Klinipath), where after approximately 6 µm thick cryosections were cut and mounted on 3-aminopropyltriethoxysilane (APES, Sigma-Aldrich) coated glass slides. After drying, the sections were fixed in acetone. Cryosections were air-dried and subsequently washed in PBS for 5 min after which they were incubated with 10% (v/v) goat serum in PBS for 30 min at 37°C to decrease background staining. Subsequently, the sections were incubated with the primary antibody HB61A (anti-CD4, VMRD Inc., Pullman, USA) or 73/6.9.1. (anti-CD8, VMRD Inc.) 1/50 diluted in PBS during 1h at 37°C. In a last step the sections were incubated with the FITC-conjugated secondary goat anti-mouse antibody (in PBS) supplemented with 1% horse serum, during 1h at 37°C. The sections were mounted in glycerol containing DABCO to counter photo bleaching. All incubations were carried out in a humidified environment, and between the incubation steps the slides were washed 3 times with PBS for 5 minutes each. Controls omitting the primary and/or the secondary antibodies were performed.In all three horses, the gross and histological lesions confirmed the diagnosis of PNE. The IH revealed important infiltrates of CD4 and CD8 positive T-cells in all three horses. In the first case more CD4 than CD8 positive T-cells were present; in the second and third case more CD8 than CD4 positive T-cells were present. The controls were negative for as well CD4 as CD8 positive T-cells.These results indicate a role for CD4 and CD8 positive T-cells in the pathogenesis of PNE. CD4 positive T-cells recognize antigen in association with MHC II and are often defined as T helper cells which stimulate B-cells and macrophages. CD8 positive T-cells recognize antigens in association with MCH I and are defined as cytotoxic T-cells. The interval between the onset of the disease and the euthanasia was clearly longer in the first case which had more CD4 positive T-cells, than in the second and third case which had more CD8 positive T-cells. So it can be stated that the longer the disease carries on, the T-cell response will change from a T-helper cell pattern into a cytotoxic T-cell pattern.
