- W2275243345 abstract "Butea frondosa is a common Indian plant. The antifertility activity of the alcoholic extract and its crystalline fraction obtained from the petals were investigated. Dried petals were kept in rectified spirit for 1 week the extract filtered then dried. The alcoholic extract of petals was washed with petroleum ether and kept at 5 degrees C. for 1 week. Crystals were then filtered out. Washed with water and recrystallized from ethanol. Melting point of crystals was 194-195 degrees C. Antifertility activity was tested in rats by administering differing doses of the alcoholic extract or the crystalline fraction dissolved or suspended in water daily for 5 days after mating. Positive control animals received 2 doses of noracyclin (Ciba). Laparatomy was done on Day 10 of pregnancy to check implants. Fetuses were delivered by Caesarian section on Day 20. In another experiment 20 rabbits received 20 mcg estradiol dipropionate in oil im; then 1 group received 200 mg/kg of the crystalline fraction by stomach tube. After 48 hours all received 1 mg of copper acetate iv and at 36 hours all were sacrificed and ovulation points in each ovary counted. Fertility was also checked in 20 male rats with doses of 25 50 and 100 mg/kg of the crystalline fraction orally daily for 10 days. After sacrifice on Day 11 weights of seminal vesicles pituitary glands and testes were noted. Action on the uterus was studied in another group of 48 mice given varying amounts of the crystalline fraction beginning 8 days after ovariectomy and continuing for 7 days. (Some of this group also received estradiol dispropionate.) The petals of Butea frondosa contain 2 glycosides butrin and palasitrin. Because of similar melting points the crystalline fraction is thought to contain a large amount of butrin. Both the crystalline fraction and the alcoholic extract reduce the number of implants in mated rats. The crystalline fraction was a fairly potent antifertility agent but not complete as a few animals did develop normal fetuses in spite of administration. Surviving fetuses showed no malformations. Noracyclin a common contraceptive on the Indian market was used for comparison; in these rats implants were found on Day 10 but were resorbed by Day 20. The crystalline fraction of Butea frondosa did not inhibit ovulation induces by copper acetate in rabbits and did not have antiovulatory gonadotrophic antiestrogenic or estrogenic action.
