- W2275688640 abstract "Glucagon-like peptide-1 (GLP-1) is an incretin and anorexigenic hormone released by the gut. GLP-1 receptor (GLP-1R) agonists are currently used in type-2 diabetes and obesity treatments. The vagal afferent neurons (VAN) express GLP-1R and have been implicated in the mediation of gut-GLP-1 effects on satiation and glycemia. Here, we study the hypothesis that vagal GLP-1R also modulates food reward behaviour. We used a rat model with a specific knockdown of GLP-1R expression in VAN (GLP-1R kd) by injecting a lentivirus carrying a shRNA sequence in the nodose ganglia. We found that GLP-1R kd rats displayed a specific short-term reduction in consumption of a novel palatable food, that was not supported by changes in innate anxiety (elevated plus maze and open field tests) or food neophobia. Moreover, in a conditioned place preference paradigm, GLP-1R kd showed a deficit in palatable food-induced side preference. Together these findings suggested that GLP-1R kd rats had a reduction in short-term hedonic appreciation of a novel palatable food. Based on the classical role of VAN GLP-1R in satiation, we hypothesized that GLP-1R kd induced compensatory mechanism at the central level that accounted for the short-term reduction in hedonic appreciation of food reward. Our research will further focus on elucidating these compensatory mechanisms. Overall, our research contributes to support the hypothesis that gut-GLP-1 mediates a signal to the brain via VAN GLP-1R that is involved in some aspects of the food reward behaviour." @default.
