It describes important strategies for generating the illumination pattern and illumination sequences. This is complemented by an analysis of requirements for fluorescent probes, dye combinations for dual-color STED, and sample preparation techniques. STED microscopy is based on the more general concept of reversible saturable optically linear fluorescence transitions (RESOLFT). The chapter explains the specifics of data acquisition as well as of data analysis and visualization, and presents a topography of application areas. A further focus is constituted by a discussion of the setup of the base instrument, the laser light sources, criteria for the choice of a suitable detector, tips for adjusting the laser power settings and scan settings, possibilities to increase the signal from a STED sample, and requirements for obtaining a high-quality point spread function (PSF). The development of novel dyes and labeling technologies is expected to boost the application range of STED." @default.
