• W2277437720 abstract "Oncidium Gower Ramsey MADS box gene 8 (OMADS8) is a PISTILLATA (PI) homologue, encoding a conserved PI motif in the C-terminal. This study focused on investigating the effects of the PI motif and C-terminal of OMADS8 protein on floral organ formation. In 35S::OMADS8 transgenic Arabidopsis, sepals were transformed into petaloid structures which showed conical-shaped epidermal cells. In 35S::OMADS8ΔPI (PI motif deleted) and 35S::OMADS8ΔC (C-terminal 29 amino acids deleted) transgenic plants, the petaloid phenotype was reduced. Further transgenic analysis by pi mutant complementation test revealed that 35S::OMADS8 rescued the petal and stamen deficiencies in the pi mutant, and produced petaloid sepals at the same time. The complementation ability of 35S::OMADS8ΔPI in the pi mutant was weaker than 35S::OMADS8, and deficiencies were only partially rescued. However, no complement phenotype was observed in 35S::OMADS8ΔC transgenic pi mutants. Thereby, OMADS8 possessed similar function with PI in regulating petal and stamen development, and this function would be abolished once its C-terminal region was removed. The effect of PI motif and C-terminal truncations on the dimerization of OMADS8 with other B functional MADS proteins was elucidated by yeast two-hybrid and FRET analysis. The result indicated that OMADS8 not only formed heterodimers with Oncidium AP3 homologous proteins OMADS3/5/9 but also with Arabidopsis AP3 to enter to the nucleus. However, the PI motif and C-terminal truncations reduced, not abolished, the dimerization of OMADS8 with AP3. In pi mutants, the expression of NAP, a direct target gene of AP3/PI complex, was up-regulated by OMADS8, irrespective of the integrity of the C-terminal sequence. The expression of GNC, BNQ1 and BNQ2, which were known to be suppressed by AP3/PI, was down-regulated by OMADS8 and OMADS8ΔPI but not by OMADS8ΔC. The expression of GNL, which was also suppressed by AP3/PI, was only down-regulated by OMADS8. Thus, the complete loss of the C-terminal sequence will abolish the gene regulation of OMADS8 and resulted in the abnormal petal and sepal development whereas the PI motif was dispensable for OMADS8 in regulating certain AP3/PI downstream genes." @default.
