- W22777729 abstract "This report extends the work of Ascher et al (1) and demonstrates that medium-dialysed human leucocyte extracts (TFDM) augment lymphocyte transformation to PPD and PHA in vitro. In this respect TFDM from Mantoux-negative individuals is just as effective as TFDM from Mantoux-positive individuals; the augmenting activity of TFDM bears a close relationship to the extent of lymphocyte DNA synthesis induced by PPD or PHA alone; and cell populations which give low responses to PPD or PHA are likewise less responsive to this augmenting factor. The augmenting activity is present in water-dialysable TF (TFW) and in preliminary experiments can be demonstrated in certain TFW fractions eluted from G25 Sephadex. In the tuberculin system the data therefore reveals an antigen-dependent but not an antigen (PPD)–specific effect of TF in vitro. It is of particular interest that dialysable extracts of non-lymphoid cell lines (L cells; BKH-21 cells), and of buffy coat leucocytes from a blood transfusion centre, also augment lymphocyte transformation to PPD. In view of the ability of TF to non-specifically increase certain aspects of the immune response in vivo, our findings using this in vitro system point to the need for identification of the augmenting factor and for clarification of its relevance to therapeutic effects of TF." @default.
