- W2277774462 abstract "This chapter addresses the adverse effects of nutraceuticals and examines the biochemical mechanisms underlying their beneficial action and toxicity. Nutraceuticals exert their activity at the systemic level by interacting with multiple targets in different organs, although the major effects are achieved through their antioxidant and anti-inflammatory activities. Phytochemicals (particularly polyphenols) represent the main group of nutraceuticals, and there is mounting evidence on the adverse effects and toxicities following the use of pharmacological doses of certain formulated products used for prevention or treatment of many common disorders. The increased use of these compounds by consumers at higher doses and chronic administration may result in adverse effects. As nutraceuticals are exempt from extensive preclinical efficacy and toxicity testing by Regulatory Agencies, their potential for harmful effects for consumers’ health cannot be ruled out. The hepatotoxicity of phytochemicals has been subjected to an Adverse Outcome Pathway approach for a better understanding of the biological plausibility of the putative mechanisms underlying this toxicity. It is envisaged that a better knowledge of the mechanisms of toxic action might be of help in the prevention and treatment of potential long-term adverse effects." @default.
