- W2282783147 abstract "A dose-related incidence of mammary nodules was observed in beagles receiving quarterly im injections of Depo-Provera (medroxyprogesterone acetate) at 1 6.1 and 23.7 times the recommended human contraceptive dosage. These dosages are approximately 60% 37 times and 14.2 times the preestablished effective dose for producing anestrus in bitches. A similar incidence of mammary nodules was observed in beagles receiving weekly im injections of progesterone at doses that are equivalent to the high and low doses of Depo-Provera. Most of these nodules were round and moderately firm. Although some mammary nodules have progressively increase in size others have disappeared spontaneously. So far there has been no evidence that any nodule was milignant although this cannot be properly evaluated until all nodules can be studied histologically when the dogs die or are sacrificed at the termination of the study. Histological examination of the mammary nodules of the dogs that have died indicated that the majority of these nodules are localized areas of nodular mammary hyperplasia. These hyperplastic nodules are associated with generalized mammary hyperplasia which was seen in all of the dogs receiving high doses of Depo-Provera or progesterone. There were 3 small benign mixed mammary tumors in 1 high-dosed Depo-Provera dog and 1 small benign mixed mammary tumor in 1 high-dosed progesterone dog. A similar incidence of hyperplastic mammary nodules and mixedmammary tumors has been induced in beagles by other progestational agents (Nelson et al. Journal of the National Cancer Institute 51: 1303 1973). Large-scale clinical studies have demonstrated that quarterly injection of medroxyprogesterone acetate to a large number of women for as long as 6 years does not produce any increase in the incidence of mammary nodules (McDaniel et al. Journal of Biosocial Science 5: 83 1973). Therefore it appears that the induction of mammary nodules in beagles by Depo-Provera is a species-specific effect that does not occur in humans.(Full text)" @default.
