- W22885736 abstract "Eyeblink classical conditioning is a useful model system for the study of the neurobiology of learning, memory, and aging which also has application to dementia. Parallel age differences in conditioning in the short-delay procedure appear in middle age in rabbits and humans that may be caused at least in part by loss of cerebellar Purkinje cells. It is only when the CS-US interval is extended to 1500 ms that the age differences in delay eyeblink classical conditioning disappear. The smaller body of evidence on the effects of normal aging in the trace, discrimination, and other complex classical conditioning procedures also suggests age-related impairment. Eyeblink classical conditioning has relevance to AD because of the involvement of the hippocampal cholinergic system. AD profoundly affects the hippocampal cholinergic system, and our observation that Alzheimer’s patients condition poorly or not at all may be related to the abnormally functioning hippocampus and loss of hippocampal CA1 pyramidal cells. Alzheimer’s patients and patients with Down’s syndrome and AD consistently perform poorly in eyeblink conditioning, but patients with dementia diagnosed as cerebrovascular dementia are not consistently poor conditioners. We suspect that CVD patients condition poorly only when some of the circuitry involved in classical conditioning has been affected. Patients with HD, PD, and Fragile X syndrome condition as well as normal age-matched adults. Eyeblink conditioning may be useful in differential diagnosis in dementia and also in detecting AD early in its onset. Because eyeblink conditioning is simple, non-threatening, and non-invasive, it may become a useful addition to test batteries designed to differentiate normal aging from dementia and AD from other types of dementia. Eyeblink conditioning may also detect AD early, before declarative memory impairment is evident." @default.
