- W2289788460 abstract "We provide evidence that a novel factor other than the lipid mobilizing factor, zinc alpha 2 glycoprotein (ZAG), promotes lipolysis in the MCA-induced sarcoma-bearing rat cachexia model. We hypothesize malignant growth promotes changes in circulating factor(s) that alter adipocyte biology prior to the clinical manifestations of cachexia. We compared tumor-bearing ad lib fed rats (TB) to non-tumor-bearing ad lib fed rats or non-tumor-bearing pair-fed rats at various time points throughout development of tumor induced cachexia. Prior to cachexia, the TB rats lost more than 10 + 0.7 % of their body fat before losing protein mass and decreasing their food intake (markers of cachexia). Fat loss occurred because adipocyte size, not number, was reduced. Increased turnover of palmitate and significantly higher serum triglyceride levels prior to cachexia were additional indicators of an early loss of lipid from the adipocytes. Circulating levels of known lipolytic hormones, norepinephrine and epinephrine, cytokines such as TNF-á, and ZAG were not increased prior to the loss of fat mass. We provide evidence that a serum factor(s) promotes the loss of stored adipose lipid prior to the loss of lean body mass in this model and that the serum factor(s) is most likely a protein that is not ZAG.
