- W2290722182 abstract "Background: Recent studies reveal platelets’ increasingly important role in inflammation in addition to thrombosis. Platelet-neutrophil adhesive interactions with endothelium have been well described; however, whether platelets alter neutrophil migration across endothelium remains unknown. Hypothesis: Activated platelets enhance neutrophil transmigration across endothelial cells. Methods: Cultured human umbilical vein endothelial cells (HUVEC) were stimulated with IL-1β 3–4 hours prior to the experiment. Neutrophils (PMN) and platelets isolated from healthy adults were injected into static adhesion chambers and recorded under phase-contrast microscopy. Adhesion or transmigration of PMN alone, PMN with unactivated platelets, and PMN with platelets activated with thrombin receptor agonist peptide (TRAP) were compared using ANOVA. Results: At 500 seconds, PMN with TRAP-activated platelets migrated significantly more than PMN alone (57±6% vs. 33±5%; p<0.01). PMN with unactivated platelets demonstrated a small increase in migration (40±4%; p<0.05) as compared to PMN alone. At 1000 seconds, there were no differences in either firm adhesion or transmigration of PMN between the groups. Conclusions: Under static conditions, activated platelets enhance early transmigration of PMN, but do not affect firm adhesion or late transmigration across IL-1β-stimulated HUVEC.
