In der Planungsphase einer derartigen klinischen Studie ist die Fallzahlplanung ein wichtiger Baustein, um das erfolgreiche Abschliesen einer Studie zu gewahrleisten. Um einen Stichprobenumfang zu bestimmen, wird zunachst eine Annahme an die Storparameter benotigt. Die Bestimmung der Grosen, die oftmals auf historischen Daten basiert, beinhaltet eine starke Unsicherheit. Aus diesem Grund hat das Studiendesign der internen Pilotstudie in den letzten Jahren stetig an Bedeutung gewonnen. Dieses erlaubt nach Rekrutierung der ersten Patienten eine ver- bzw. entblindete Schatzung der Streuung, so dass die Fallzahl hinsichtlich ihrer initialen Annahme uberpruft wird. In der vorliegenden Arbeit werden zunachst bestehende Verfahren zur Fallzahlanpassung analytisch sowie in Simulationsstudien untersucht und verglichen. Eine Erweiterung der Methodik zur Fallzahlanpassung bei zeitvariierenden Ereigniszeitdaten bildet den Mittelpunkt der Dissertation." @default.
