- W22967069 abstract "Four aflatoxin ? 1 (AFB 1 ) derivatives (I–IV) separable by high pressure liquid chromatography were formed by mild acid hydrolysis of the DNA– or RNA–AFB 1 adducts formed in rat liver in vivo or in vitro by rodent liver icrosomes supplemented with AFB 1 , NADPH, and DNA or RNA. Product II was 2,3–dihydro–2,3–dihydroxy-AFB 1 (the dihydrodiol), which was previously demonstrated in this laboratory as a hydrolysis product of the adducts. On the basis of chemical and radiochemical data, the hydrolysis of I, III and IV to the dihydrodiol, the conversion of I to III, and the interconversion of I and IV, each of these three products was deduced to result from N–7 substitution of guanyl residues by carbon–2 of AFB 1 –2,3–oxide. The properties of the products were entirely and uniquely consistent with the following structures: I, 2,3–dihydro–2–(N 5 –formyl–2,5,6–triamino–4–oxo–pyrimidin-N 5 –yl)–3–hydroxy–aflatoxin B 1 ; III, 2,3–dihydro–2–(guan–7-yl)–3–hydroxy-aflatoxin B 1 ; IV, 2,3–dihydro–2–(8,9–dihydro–8–hydroxy–guan–7–yl)–3–hydroxy-aflatoxin B 1" @default.
