- W2298012368 abstract "In this chapter, we will briefly describe the functioning of the hypothalamic–pituitary–adrenal (HPA) axis and the possible role of its genetic variations in smoking behaviors. In particular, we will characterize on a molecular level the structure and mode of action of glucocorticoid (GR) and mineralocorticoid (MR) receptors, discussing association findings, from candidate gene and genome-wide association studies, involving the NR3C1 and NR3C2 genes (encoding the GR and MR, respectively) and smoking. The HPA axis has a putative role in various aspects of smoking behaviors and there is evidence that GRs and MRs are involved in pathophysiological processes related to addiction. Additionally, an MR/GR balance hypothesis suggests that an imbalance between these receptors could result in dysfunctional stress responses, increasing susceptibility to stress-related diseases, including smoking behaviors. Thus, functional polymorphisms in the NR3C1 and NR3C2 genes can be considered strong candidates for genetic association studies of smoking." @default.
