- W2298991168 abstract "This chapter outlines the historical and modern efforts dedicated to examining the “premature aging hypothesis” of substance abuse. This hypothesis has been tested primarily using neuropsychological studies, which tend to generate inconsistent findings. Modern advances in the identification of aging biomarkers and neuroimaging technologies have sparked renewed interest in the premature aging phenomenon. Cellular senescence was identified to be a key biochemical process that underlies the aging process, and telomerase activity, which reflects cellular senescence, was also substantially reduced in a group of heroin abusers. Moreover, telomerase activity can be modulated by heroin abuse to influence brain systems that are closely related to aging, including the prefrontal cortex and medial temporal lobe. Therefore, heroin-associated accelerated aging at the cellular level was related to accelerated aging at the brain system level. These findings contributed to understanding the potential mechanisms underlying the anecdotal evidence that substance abusers appear older than their actual age. The results also set the stage for future studies to push beyond the existing boundaries of the understanding of addiction pathology." @default.
