- W2300327424 abstract "SNARE proteins are involved in the fusion of transport vesicles with target membranes in all eukaryotes and form complexes in which 3 Q-SNAREs interact with one R-SNARE. In the first part of the work I tried to find additional proteins which interact with the yeast Q-SNARE Vti1p in the transport from the trans-Golgi to the prevacuole and vacuole via an multicopy suppressor screen. I was able to identify a previously uncharacterized non-essential gene, which was named VTS1, and the R-SNARE YKT6 both as suppressors of the growth and vacuolar transport defect of the temperature sensitive vti1-2 mutant. VTS1 showed a specific genetic interaction with VTI1 only in the last transport step to the vacuole, but not in the transport to the prevacuole. In addition to its previously described function in retrograde traffic to the cis-Golgi and in homotypic vacuolar fusion, YKT6 also interacted with VTI1 in traffic to the prevacuole and vacuole. This indicates, that Ykt6p is the missing R-SNARE in the SNARE-complexes of the biosynthetic transport to the prevacuole (Ykt6p, Vti1p, Pep12p and Syn8p) and vacuole (Ykt6p, Vti1p, Vam3p and Vam7p). In the second part of the work I was able to identify and characterize a new Q-SNARE protein, named Use1p, in Saccharomyces cerevisiae via a database search, which was localized to the ER. In specific transport assays with temperature sensitive mutants of the protein I could show that Use1p is involved in the retrograde transport from the cis-Golgi to the ER. This was also supported by strong genetic interactions between USE1 and the genes encoding SNAREs in retrograde traffic to the ER. In addition, anti-Use1p-antibodies specifically co-immunoprecipitated SNAREs of the retrograde Golgi-to-ER-SNARE complex but not those of the anterograde SNARE complex. Therefore it seems, that the SNARE complex of the retrograde transport from the cis-Golgi to the ER consists of the SNARE proteins Use1p, Ufe1p, Sec22p and Sec20p." @default.
