Hybridomas were selected byspecific immunoassay reaction oftheir fluid withcrudeyeast killer toxin extract. Amongthe monoclonal antibodies, whichwerecharacterized bytheWestern blottechnique, one(designated KT4)proved tohaveprecipitating properties, thuspermitting theneutralization ofthekiller activity ofthetoxin. Experiments indoubleimmunodiffusion showedthatmonoclonal antibody KT4 produced homologous precipitin bandsbyreacting witheither thecrudetoxin usedasimmunogen oratoxic extract ofHansenula mrakii. Itissuggested that these monoclonal antibodies will beuseful forthepurification, characterization, and understanding ofthebioactions ofyeast killer toxins. Previous studies haveshownthatkiller yeasts andtheir toxins might beeffectively usedtodifferentiate strains of opportunistic yeasts forepidemiological purposes (3,4,6). Later, weobserved that thekiller phenomenon, whichwas previously considered toberestricted toyeasts, wasalso displayed bykiller yeasts against unrelated microorganisms (bacteria, hyphomycetes, lipophilic yeasts, aerobic actinomycetes, andachlorophyllous microorganisms) (9). Thediverse biological activity ofkiller yeasts andthepotential therapeuticeffect shownbya crudetoxic extract ofHansenula anomala topically applied tolaboratory animals experimentally infected withMalassezia furfur andMalassezia pachydermatis (L.Polonelli, R.Lorenzini, F.DeBernardis, andG.Morace, Mycopathologia, Myco172) prompted usto trytoisolate andpurify yeastkiller toxins.
