- W2315622012 abstract "Background: Primary effusion lymphoma (PEL). also referred to as body cavity based lymphoma, is a recently recognized lymphoma most commonly occurring in acquired immunodeficiency syndrome (AIDS) patients. PEL is a large B-cell immunoblastic lymphoma with involvement of the body eavities and commonly associated with recently identified human herpesvirus-8 (HHV-8) and EBV. Interleukin(IL)-10 is a pleiotropic cytokine with potent B-lymphocyte activation properties which has been shown to be an autocrine growth factor for other AIDS related B-cell lymphomas. Methods: We measured IL-6, IL-10 and Interferon-γ production in the tissue supernatants of the previously characterized BCBL cell lines BC-1, BC-2, BC-3(HHV-8+,EBV+) and BCBLI(HHV-8+.EBV-) by enzyme linked immunoabsorbent assay(ELISA). The role for IL-10 as a growth factor was determined by tritiated-thymidine incorporation following treatment with various concentration of blocking anti-IL-10 antibody and antisense IL-10 oligonucleotides. High grade B cell lines with and without IL-10 production were also tested as a control. IL-10 transcript levels of PEL, cell lines were also determined by northern blot analysis using a radioactively labelled IL-10 specific oligonucleotide probe. Intracytoplasmic IL-10 was detected by flow cytometric analysis. Results: Supernatants from all in vitro cultured PEL cell lines showed very high levels of IL-10 (range:5000-15000 pg/ml) by ELISA while IL-6 production was minimal. No Inf-γ secretion was detected. All of the PEL cell lines showed intracytoplasmic IL-10 by flow cytometric analysis and high levels of IL-10 mRNA by northern blot analysis. Both anti-IL-10 neutralizing antibody and IL-10 antisense oligonucleotides significantly inhibited PEL cell line proliferation while blocking anti-IL-6 antibody showed no significant inhibition. Conclusion: These findings support the hypothesis that IL-10 is an autocrine growth factor in PELs. IL-10 appears to play an important role in the pathogenesis of PELs. similar to some of the high grade AIDS associated B cell lymphomas. These data also suggest that inhibition of PEL growth by IL-10 could serve as a potential therapeutic strategy for the treatment of PELs." @default.
