The compounds which have the ability to induce immunomodulatory effects on DCs may be employed for the treatment of autoimmune diseases and allograft rejection. The aim of this study was to investigate the in-vivo effects of 18α-Glycyrrhetinic acid on DCs. 10 milligram/kg 18α-Glycyrrhetinic acid was injected intra-peritoneally into BALB/c mice every other day within 4 weeks, and splenic DCs were isolated using nycodenz. The phenotypic and functional properties of DCs were studied by flow cytometry and mixed lymphocyte reaction (MLR), respectively. In addition, the percentage of Tregs was compared in both groups and the level of IL-4 and IFN-γ in MLR supernatant and mice serum were measured by ELISA kits. Determination of TLR mRNA expression level using real time PCR, other accessory of this study, will be done. As a result, flow cytometry analysis showed expression of CD40 (P<0.0250) and MHC II (P<0.0048), as maturation markers and costimulatory molecules were significantly decreased. Also, expression of CD86 was not changed in test group compared to control group. The percentage of Tregs in control (2.350±0.8227) and test (1.276±0.4477) groups was not statically significant. The level of IL-4 and IFN-γ was not different in MLR supernatant and serum of both groups. Allogeneic T cell stimulation in MLR was also significantly inhibited in comparison with the control groups (p<0.05). Our data indicate that in-vivo 18α-Glycyrrhetinic acid administration inhibits maturation and activation of DCs in the same manner as in-vitro conditions.
