- W2317258981 abstract "Purpose: The global shortage of human (h) donor corneas limits clinical corneal transplantation (Tx). The genetically-engineered (GE) pig (p) may provide an alternative source for both corneal keratoplasty and cultured corneal endothelial cell (CEC) Tx. The aim of this study was to compare density, morphology, and in vitro proliferative capacity of CECs from wild-type (WT) and GE pigs and humans. Methods: Density and size of CECs, and the percentage of hexagonal cells (as a measure of heterogeneity) were measured by confocal microscopy in corneas from WT and GE pigs of different ages -neonatal (4-5d), young (8-15w), middle (5-15m), and old (20-25m). The proliferative capacity of cultured CECs was measured by direct cell counting and BrdU assay. hCECs from donors Â≤36 years (young) and ≤49 years (old) were also used in this study. GE pigs included CD46 and GTKO/CD46/CD55 (which protect from the human immune response). Results: Mean CEC densities (cells/mm2) were neonatal (5,968), young (3,789), middle (2,589) and old (2,070). As with human corneas, there was an age-dependent decrease in pCEC density and increase in pCEC size. However, unlike human corneas, there was no correlation between the percentage of hexagonal cells and age (approximately 50% in all pig corneas vs >60% in young and < 50% in old human corneas), suggesting that heterogeneity is intrinsic for pig corneas. Both young pig and human CECs had a significantly higher proliferative capacity than old CECs (p< 0.01). However, during culture there was a significantly higher percentage of apoptosis in hCECs compared to pCECs (p< 0.01). Genetic modification did not affect pCEC density, morphology, proliferative capacity, or cell viability. Conclusion: Because of the availability of young pigs, greater CEC density, excellent proliferative capacity of cultured pCECs, and reduced cell death during culture, GE pigs could provide an unlimited source of corneas and CECs for clinical Tx." @default.
