- W2317261390 abstract "Under the various simplified methods of determination of CO-Hb concentration in blood which have been used up to the present, the measurement has to be made instantly after blood letting, or the blood has to be isolated from air by paraffin, as CO is easily dissociated from Co-Hb. These are some difficulties with the various simplified methods of the determination of CO-Hb at the actual place the air of which contains the gas. As the dissociation of CO from CO-Hb was due to its contact with air, a new method using a melangeur for leucocytes in blood letting was tried to prevent the dissociation by isolating blood from the air thus making CO-Hb stable and the immediate measurement no necessary. As this new method is characterized by easy application, by keeping blood in the same condition for some hours, and by portability, it can be incorporated into any method of CO-Hb determination. On the other hand, now that spectrophotometric technic has been established and popularized, the making use of this technic seems the best method of the determination. And, therefore, spectrophotometric determination of CO-Hb concentration in blood using a melangeur for leucocytes was studied. In this spectrophotometric determination of CO-Hb, the difference of the absorbancy-curves between CO-Hb and O2-Hb or red-Hb is generally utilized, and we drew these three curves in the visible wave-length region. In consequence of this study, the ratio of the absorbancy of CO-Hb to that of red-Hb at 419 mμ was found to be applied. The blood of a rabbit was used in our experiments, and CO-Hb was saturated by experimentally generated CO gas. Red-Hb was reduced by Na2S2O4, and O2-Hb was diluted by Na2CO3. First of all, the stability of CO-Hb in a melangeur was tested. And then, experiments on the stability of CO-Hb in a melangeur were carried out on the fellowing two kinds of blood, (1) 25 and 50% CO-Hb solution experimentally prepared and sucked into a melangeur, and (2) the blood of rabbits, which actually inhaled CO gas, sucked into a melangeur. Changes of absorbancies of those Co-Hb solutions were measured three times in both experiments, i.e. instantly, after 3 hours and after 6 hours. Owing to the result that these absorbancies in both experiments did not decrease, it was concluded that Co-Hb in a melangeur was stable for 6 hours. Next, the relationship between the ratio of the absorbancy of every concentration of Co-Hb solutions to that of saturated Co-Hb solution at 419 mμ and the Co-Hb concentration were studied, and its calibration-line was drawn. This line appeared straight, and its empirical formula was, y=0.00552x-0.5136 (x:Co-Hb%, y:the ratio of the absorbancy of every concentration of Co-Hb solutions to that of saturated Co-Hb solution at 419mμ) The error of this line was about 3.1% of Co-Hb concentration in the blood, hence the error of this method was not as grave as that of common simplified methods. Now, our actual use this simplified method of spectrophotometric determination of Co-Hb concentration in blood by the use of a melangeur for leucocytes was as follows. Blood was let out of an earlobe into the melangeur to the scale I, and instantly after that 0.1% Na2CO3 solution was sucked into the scale II, and the melangeur was shaken and capped with a rubber band. Blood was allowed to remain in this condition for 6 hours. At the spectrophotometric measurement, the contents of the melangeur was put into a small test-tube, and 0.1 cc of it was taken into pipette, and was diluted to 1:2, 000 with 0.5% Na2CO3 and 0.5% Na2S2O4 solutions, and the absorbancy of this solution A was measured at 419mμ. On the other hand, the remaining solution was saturated with coal gas, and the absorbancy of this solution B was measured likewise. The ratio between the both absorbancies A/B was calculated. By this ratio and the above mentioned calibration-formula, Co-Hb concentration in the blood was determined." @default.
