- W2317291505 abstract "Chronic itch is a debilitating symptom of many diseases and dry skin, or xerosis, can be a contributing factor. However, the neurophysiological changes associated with dry skin are still poorly understood. The rodent cheek behavioral model is able to reliably distinguish between itch- and pain-producing stimuli. Mice scratch with the hind-limb when pruritogens are injected into the cheek, but wipe with the forelimb when algogens are applied. We used the acetone-ether-water (AEW) model to induce dry skin at the cheek. Spontaneous scratching was significantly increased compared to the water-only control, while there were no changes in wiping. Immunohistochemical studies of the cheek skin demonstrated that total intraepidermal fiber density was significantly increased in the dry skin group. This increase was present even with the use of Elizabethan collars which prevented scratching, indicating that the dry skin itself was capable of enhancing intraepidermal fiber density. We further characterized the dry skin-induced hyperinnervation using peptidergic (CGRP) and non-peptidergic (Ret) markers. Ret+ fibers were significantly increased after AEW treatment, but CGRP+ fibers were not different, suggesting that Ret+ fibers may be important for dry skin-induced pruritus. Calcium imaging studies of dissociated trigeminal neurons from treated mice demonstrated an increase in cells responsive to chloroquine, a non-histaminergic pruritogen.
