Donadi1 1 National Institute of Science and Technology in Stem Cells and Cell Therapy, Brazil Mesenchymal stromal cells (MSCs) have immunosuppressive effects and have been used for the treatment of autoimmune diseases. We evaluated the gene expression profiles and the in vitro immunommodulatory properties of MSCs obtained from multiple sclerosis patients and controls. Bone-marrow MSCs were isolated by plastic adherence. Gene expression profile was evaluated by microarrays by using Agilent platform. For coculture assays, CFSE-labeled PBMCs were added to patient or control MSCs, and stimulated with PHA. After five days, T-cell proliferation was assessed by flow citometry. IL-10 and TGF-beta were measured in supernatants by CBA flex. The results were analysed by the Limma-T and Mann-whitney tests. The study was approved by the local Ethics Commitee. Compared to controls, we found 641 differentially expressed genes in patient MSC (P<0.01; fold change>2), comprising 262 repressed genes, including TGFB1 and HGF and, 379 induced, among them, IL10 and IL6. The inhibitory effect of patients MSCs (52.1 + 5.6%) on T-cell proliferation was significantly reduced (P=0.024) compared with controls (69.4 + 7.2%). There were no differences in the percentages of CD4+CD25hiFoxp3+ cells in both MSCs cocultures. IL-10 and TGF-beta secretion were significantly diminished (P<0.05) in supernatants from cocultures with patient MSCs (IL-10: 53.8 + 49.4; TGF-beta: 59.4 + 24.6 pg/mL) compared with controls (IL-10: 109.9 + 64.6; TGF-beta: 226.6 + 153.2 pg/mL). Concluding, patient and control MSCs exhbited distinct transcriptional profiles and patient MSCs secreted low amounts of IL-10 and TGF-beta and exhbited decreased ability to inhibit T lymphocyte proliferation.
